J Neurophysiol 87: 3117–3125, 2002; 10.1152/jn.00745.2001. We used anatomical and physiological approaches to characterize nicotinic receptors (AChRs) on Renshaw cells of the neonatal rat spinal cord. Confocal imaging of Renshaw cells, identified by their characteristic pattern of gephyrin immunoreactivity, revealed that these neurons are immuno-positive for the 4 and 2 AChR subunits but not for the 7 subunit. We used whole cell recording in spinal cord slices to characterize synaptic transmission from -motor neurons to Renshaw cells, which could be identified pharmacologically by the sensitivity of transmission to d-tubocurarine. -Motor neuron-to-Renshaw cell synapses were blocked by 10 M dihydro-erythroidine (dH E), but not 50 nM methyllycaconitine (MLA), a selective 7 antagonist. These findings support a role for 4 2-like AChRs, but not 7 AChRs, in rapid excitatory transmission between -motor neurons and Renshaw cells in rat spinal cord.

متن کاملInhibitory synaptic modulation of renshaw cell activity in the lumbar spinal cord of neonatal mice.
In the mammalian spinal cord, Renshaw cells (RCs) are excited by axon collaterals of motoneurons (MNs), and in turn, provide recurrent inhibition of MNs. They are considered an important element in controlling the motor output. However, how RCs are modulated by spinal circuits during motor behaviors remains unclear. In this study, the physiological nature of inhibitory synaptic inputs to RCs in...
متن کاملActivity of Renshaw cells during locomotor-like rhythmic activity in the isolated spinal cord of neonatal mice.
In the present study, we examine the activity patterns of and synaptic inputs to Renshaw cells (RCs) during fictive locomotion in the newborn mouse using visually guided recordings from GABAergic cells expressing glutamic acid decarboxylase 67-green fluorescent protein (GFP). Among the GFP-positive neurons in the lumbar ventral horn, RCs were uniquely identified as receiving ventral root-evoked...

متن کاملShort- and long-term enhancement of excitatory transmission in the spinal cord dorsal horn by nicotinic acetylcholine receptors.
Spinal administration of nicotinic agonists can produce both hyperalgesic and analgesic effects in vivo. The cellular mechanisms underlying these behavioral phenomena are not understood. As a possible explanation for nicotinic hyperalgesia, we tested whether nicotinic acetylcholine receptors (nAChRs) could enhance excitatory transmission onto spinal cord dorsal horn neurons. Whole-cell patch-cl...
